Among principle antioxidative enzymes, SOD1 and SOD2 are reportedly decreased in trinitrobenzenesulfonic acid-induced colitis model rats [46]. However, our data on DSS-induced colitis model mice showed no significant changes in the levels of SOD1 and SOD2 but rather indicated a prominent decrease in the levels of GPX1 and GPX2 (Fig. 4), which might also be associated with the aggravated intestinal damage.

Most of the current therapies for IBD involve treatment with glucocorticoids, 5-aminosalicyclic acid and immunosuppressive drugs, but these are not safe as they may produce serious complications and toxic side effects such as nephrotoxicity, pulmonary toxicity and male infertility [3,6,7].

In previous experiments, we found that melatonin 1, the main compound secreted by the pineal gland, inhibits the rat striatum nNOS activity in a dose-dependent manner [20]. On the other hand, in a further paper, it was found that melatonin acts as non-competitive inhibitors of nNOS, being calmodulin the target involved in the nNOS inhibition activity of this compound [21].

2009, European Journal of Medicinal ChemistryCitation Excerpt :Our search group has reported several nNOS inhibitors with a kynurenine structure 2, showing a significant nNOS inhibition activity [34,35]. A second type of nNOS inhibitors described by our research group show a kinurenamine structure 3 [36] among them the main melatonin brain metabolite (AMK: 3, R1 = MeO, R2 = Me). All these compounds inhibit nNOS in a dose-dependent manner, and it has been found that AMK rather than melatonin is the active metabolite against nNOS in rat striatum [37].
