ESR Spin Trapping Investigation on Peroxynitrite Decomposition: No Evidence for Hydroxyl Radical Production

Abstract

The decomposition of peroxynitrite in the presence of 5,5-dimethyl-1-pyrroline N-oxide (DMPO) generated 5,5-dimethylpyrrolidone-(2)-oxy-(1) (DMPOX) without formation of DMPO/·OH. Formate enhanced the peroxynitrite decomposition but did not generate any detectable amount of formate-derived free radicals. Glutathione, cysteine, penicillamine, and ascorbate reacted with peroxynitrite to generate the corresponding thiyl and ascorbyl radicals. The results show that the decomposition of peroxynitrite did not generate any significant amount of ·OH radicals, and one-electron reduction of peroxynitrite by ascorbate may be one of the important peroxynitrite detoxification pathways.

    During discharge, an obvious signal appeared, except for the fourfold peak of conventional DMPO-OH (1:2:2:1), the triplet in the EPR spectrum belonging to 5,5-dimethyl-2-oxopyrroline-1-oxyl (DMPOX). Active substances such as RNS and O2 (a1Δ) existing in the discharge system interact with DMPO to form C(OO•)-N•(O), and the double free radical will rearrange into strongly oxidizing nitrogen-like groups and generate DMPO-O2H and DMPOX after a redox reaction with DMPO (Bilski et al., 1996; Liu et al., 2016; Shi et al., 1994). In addition, nitro radicals or peroxynitrite affect the signal peaks, causing interference with the signals of DMPO-OH and DMPO-SO4 (Wang et al., 2018).

    The ROS generation decreased when catalase, which is a specific antioxidant against H2O2, was added to cadmium-treated BEAS-2BR cells (Fig. 1A). We also measured the oxygen radical levels in cadmium-treated BEAS-2BR cells and the possible inhibition by an antioxidant using ESR spin trapping according to the method described previously (Shi et al., 1994a; Shi et al., 1994b). This method involves the addition-type reaction of a short-lived radical with a paramagnetic compound (spin trap) to form a relatively long-lived free radical product (spin adduct), which can then be studied using ESR.

    Similar results were found in a previous study (Ratnayake Bandara et al., 1992), and the signal was attributed to the 2-(N-chloroimino)-5,5-dimethylpyrrolidine-l-oxyl generated via the reaction between the nitroxyl radical and DMPO. Shi et al. (1994) observed similar electron spin resonance spectra and attributed it to the signals of 5,5-dimethylpyrrolidine-2-oxyl generated via the reaction between peroxynitrite and DMPO. However, a DMPO–OH signal generated from OH and DMPO was not observed, because the OH concentration was too low to generate DMPO-OH or the generated DMPO–OH at a low concentration was destroyed by nitroxyl radicals or peroxynitrite.

    We used DMPO and 4-POBN as spin traps to detect the coumarin free radical intermediates produced by MPO. The peroxidase-catalyzed oxidation of the hydroxylated coumarins (1 or 2) in the presence of DMPO gave the typical nine-line spectrum of 5,5-dimethyl-1-pyrrolidone-2-oxyl-1 (DMPOX), whose characteristic hyperfine coupling constants aN = 7.2 G and aH = 4.1 G, appeared at g = 2.0085, agreeing with literature values [39–41] and the corresponding simulated spectra (Fig. 5A). The acetoxylated coumarins (3 or 4) did not produce a significant EPR signal in the presence of HRP-H2O2 and DMPO under the assessed conditions.
