It has been shown that T cells, particularly those bearing γδ receptors, play a pivotal role in mucosal tissue homeostasis and immunosurveillance against invading pathogens and epithelial stresses such as malignant transformations [21–25]. γδ T cells develop mainly in the thymus and generate their γδ T cell receptor through V(D)J recombination [26–29].

Consistent with their distinct function, DETCs do not express the αβ T cell coreceptors CD4 and CD8, or the costimulatory molecules CD28 and ICOS, which are essential for αβ T cell function (Boismenu and Havran, 1998; Haas et al., 1993; Jameson et al., 2004; Shires et al., 2001). Instead, the Junctional Adhesion Molecule-Like protein, JAML (Moog-Lutz et al., 2003), was identified as the first costimulatory receptor specific to epithelial γδ T cells (Witherden et al., 2010).

While it is still not known why two T cell lineages have been conserved in all jawed vertebrates, years of study demonstrate that γδ T cells recognize antigens differently than αβ T cells, acquire effector functions faster than αβ T cells, and play specialized roles in immunity (reviewed in Refs. [1–6]). However, despite this knowledge, the signaling properties of the γδ T cell receptor (TCR) remain poorly understood.
