MAP kinase-mediated signalling to nucleosomes and immediate-early gene induction

Abstract

Extracellular signals are transduced into the nucleus through a variety of signalling mechanisms to elicit changes in patterns of gene expression. This review is focused in the MAP kinase cascades and the part they play in the induction of the immediate-early (IE) genes. We discuss the MAP kinases and their downstream effectors that are known to phosphorylate substrates in the nucleus. In addition to phosphorylating specific transcription factors, MAP kinases and their downstream kinases are inplicated in eliciting rapidly targeted alterations in the choromatin environment of specific genes by modulating the phosphorylation and/or acetylation of nucleosomal and chromatin proteins.

    In 1991, Mahadevan et al. showed that the stimulation of quiescent mouse fibroblast cells with growth factors and mitogens resulted in the phosphorylation of H3 at multiple serine sites, and ultimately the induction of immediate-early genes, many of which are proto-oncogenes whose sustained expression can have profound effects on cellular growth (Mahadevan et al., 1991) (Fig. 1). The induction of these genes, which include Fos, Fosl1, Jun, c-Myc and EGR1, was observed to be rapid and transient without requiring protein synthesis for their expression (Bahrami and Drablos, 2016; Mahadevan et al., 1991; Thomson et al., 1999). The correlation between immediate-early gene expression and H3 phosphorylation led to a consensus that these two events are mechanistically linked and was termed the ‘nucleosomal response’ (Fig. 1).
