Zusammenfassung
Mit Hilfe pharmakologischer, biochemischer und morphologischer Methoden gelang es, ein auffallendes Adaptationsphänomen zu beobachten, bei dem eine Enzymvermehrung auch morphologisch zum Ausdruck kam. Vorbehandlung mit Phenobarbital (Luminal) erhöhte die Aktivität verschiedener, nur in den Lebermikrosomen lokalisierter Fermente, die an der Oxydation und Hydrolyse zahlreicher Pharmaka beteiligt sind. Ihre Wirkungsdauer wird durch den beschleunigten Abbau beträchtlich verkürzt. Die Aktivitätssteigerung der Fermente beruht auf einer Enzyminduktion und ist verknüpft mit einer Proteinvermehrung in der Mikrosomenfraktion der Leberzellen, die auch morphologisch in einer Neubildung von Membranen desjenigen endoplasmatischen Reticulums zum Ausdruck kommt, welches frei von Ribosomen ist. Dabei verschwinden durch Bleihydroxyd darstellbare, elektronenoptisch sehr dichte Strukturen, die über das ganze Zellplasma verstreut sind und für Glykogen gehalten werden. An den mit Ribosomen besetzten Membranen des endoplasmatischen Reticulums ist keine deutliche Änderung sichtbar. Freie Ribosomenpartikel dagegen vermehren sich. Alle Strukturänderungen spielen sich vorwiegend in den Zellen der Läppchenperipherie ab. Die auffallende Oberflächenvergrößerung des endoplasmatischen Reticulums könnte für intracelluläre Transportvorgänge von Bedeutung sein.
