2022, Pharmacology Biochemistry and BehaviorCitation Excerpt :Since the CP is inhibited by γ-aminobutyric acid(GABA)ergic mircrocircuits (Groves, 1983), it may also be hypothesized that WAY-100635 disrupted the inhibition exerted by the GABAergic microcircuits in the CP, which at least may have added to the increase of available DA. In the CP, DA concentrations are regulated by autoreceptors of the D2R subtype localized at the presynaptic terminal (Langer, 1974). Remarkably, the WAY-100635-induced elevation of available DA appeared to be so high, that the inhibitory feedback mechanism of D2 autoreceptors was not effective in normalizing synaptic DA levels in the CP, at least in the present time window between administration of the antagonist and D2/3R imaging at 75 min post-challenge.

2022, Journal of Psychiatric ResearchCitation Excerpt :Schildkraut (in Hardy et al., 1986) claimed in his catecholamine hypothesis an increased noradrenergic concentration during a manic state. Next, clonidine is an antihypertensive stimulating the presynaptic alpha2 central adrenergic receptor, and as such, clonidine reduces the release of noradrenaline on the synaptic cleft, and down-regulates cellular firing rates (Langer, 1974). Further, recurrent axonal and dendritic collaterals stimulate alpha2 receptors, which branch back to inhibit the firing of their own cells (Roehrich and Gold, 1987).
