The 5-HT4 receptor family, that comprises multiple isoforms (5-HT4A, 5-HT4B, 5-HT4C, 5-HT4D, 5-HT4E, 5-HT4F, 5-HT4G, 5-HT4H, 5-HT4HB), is coupled to Gs to enhance cAMP production (Gehlert et al., 1991). The 5-HT5 receptor family comprises the 5-HT5A and the 5-HT5B receptors which have 70–88% overall sequence homology.

Their presence and abundance in these nuclei was, prior to this study, ambiguous owing to different authors reporting different patterns of expression in these ROIs depending on the methods used, as well as a lack of data on cell counts (Barnes and Sharp, 1999; Bloom and Morales, 1998; Gehlert et al., 1991; Kilpatrick et al., 1987, 1988; Laporte et al., 1992; Morales et al., 1998; Pratt et al., 1990). We found a lack of 5-HT3A receptor expression at the protein level in the SNc, at least when IHC is employed as the detection technique.
