Abstract

The subcellular localization of norepinephrine and some of its derivatives was determined in the rat brain after their intraventricular administration. The catecholamine derivatives, norepinephrine, epinephrine and dopamine, were highly localized in the synoptosomal fraction. The methoxyamine metabolites were also localized in synaptosomes but to a lesser extent. Octopamine, the β-hydroxylated metabolite of tyramine was more highly concentrated by the synaptosomal fraction than tyramine itself.

An attempt is made to unify the pathophysiology of the cerebral, cardiovascular, and renal complications of hepatic failure by postulating the accumulation of false neurochemical transmitters in the presynaptic storage sites of the peripheral and central adrenergic nervous system. Such an hypothesis is consistent with many widely accepted clinical and biochemical observations previously made about hepatic failure, hepatic coma, and the hepatorenal syndrome. It may explain the recently described beneficial effects of L-dopa on the mental and circulatory status of patients in hepatic failure and the occasionally successful treatment of the hepatorenal syndrome with large doses of metaraminol or other alpha-adrenergic amines. Experimental findings obtained in both laboratory animals and patients are presented.
Compounds antagonistic to norepinephrine retention by rat brain homogenates

Several compounds, either catechols, or amines with a phenyl-alkyi configuration or an approximation of it, diminished the ability of rat brain homogenates to retain in vitro, [³H]norepinephrine accumulated in brain tissues in vivo following intracisternal injection. Among the active compounds were several phenethylamine derivatives, including β-phenethylamine, β-phenylthanolamine, amphetamine, p-tyramine, and octopamine: compounds which are indirectly acting amines or putative false transmitters. Presence of a β-hydroxyl group may increase activity somewhat. An aliphatic side chain of 2 or 3 carbons was important for activity of aromatic amines. Also active were serotonin, several catecholamines, and 6-hydroxy-dopamine, as well as the catechol amino acid, l-dopa. Deaminated catechols, including dihydroxymandelic acid and dihydroxyphenylacetic acid were active at higher concentrations only. Catechols were inactivated by O-methylation. Certain non-aromatic cyclic amines, including amantadine, were active. Aliphatic amines and Ω-hydroxyl-amines of appropriate carbon chain length had some activity. Acetylcholine (with eserine) was inactive. That the active compounds may have significant central nervous system actions based partially on their ability to interfere with catecholamine re-uptake or storage, is suggested.
Comparative effects of p-chloroamphetamine and amphetamine on metabolism and in vivo release of <sup>3</sup>H-norepinephrine in the hypothalamus

Following the implantations of a push-pull cannula into the anterior hypothalamus and a polyethylene cannula into the lateral ventricle of the rat, the brain norepinephrine (NE) stores were labeled by intraventricular injection of ³H-NE. In freely moving nonanesthetized control animals, 4.91 + 0.73 nCi/hr of radioactivity were spontaneously released into the perfusate of the hypothalamus, with 25 percent of the total radioactivity being unchanged NE. Although amphetamine and p-chloroamphetamine caused similar qualitative alterations in the metabolic pattern of ³H-NE in brain tissue, p-chloroamphetamine evoked a striking increase in the release of NE and normetanephrine into the perfusate while pharmacologically equipotent doses of amphetamine elicited only a small increase in the release of NE; the changes in the release of normetanephrine were less pronounced than those caused by p-chloroamphetamine. Since both NE and its O-methylated product are derived mainly from stored tritiated NE, the results support the view that some of the central actions of p-chloroamphetamine may be mediated predominantly through the release of stored catecholamines while the storage of NE and dopamine appears not to be as essential for the central stimulatory action of amphetamine as long as the synthesis of catecholamines is maintained.
