In the classical model, upon fixation of its ligand and dimerization, ERα regulates gene transcription by a direct binding to DNA on ERE motif (Estrogen Response Element) (Seiler-Tuyns et al., 1986; Umesono and Evans, 1989; Green and Chambon, 1987; Kumar and Chambon, 1988; McKenna and O'Malley, 2002) and reviewed in (Klinge, 2001) (Fig. 2). This palindromic sequence “AGGTCAnnnTGACCT” is spaced by 3 nucleotides which modulate ERα selectivity and affinity for DNA (Shu et al., 2010; Anolik et al., 1993).
