Affinity of SP to NK1 receptors is about hundred-fold than to the NK2 or NK3 receptors (Guard and Watson, 1991), however, actions of SP on these two latter receptors have to be taken into consideration as well. NK2 receptors are located mainly in the thalamus, hippocampus, infralimbic and prelimbic cortex (Saffroy et al., 2003).

The NK1R is the high-affinity receptor for SP, while NK2R and NK3R bind with lower affinity to this peptide (Guard and Watson, 1991; Lomax et al., 1998; Shimizu et al., 2008). Nevertheless, SP can interact with all tachykinin receptor types with the ability to stimulate and/or inhibit propulsive motility (Holzer, 1997; Shimizu et al., 2008).

The rank order potency of tachykinins is SP > NKA > NKB for the NK1 receptor, NKA > NKB > SP for the NK2 receptor and NKB > NKA > SP for the NK3 receptor (Guard and Watson, 1991, Maggi, 1995). The structural unit of NK1 receptor is formed by seven hydrophobic transmembrane domains, extracellular and intracellular loops.

They can interact with multiple G proteins, which couple to phospholipase C via Gq and to adenyl cyclase via Gs, and thus stimulate phosphatidylinositol hydrolysis and cyclic AMP formation, respectively (Guard and Watson, 1991; Nakajima et al., 1992; Nakanishi et al., 1993). In NK1R transfected Chinese hamster ovary (CHO) cells, SP-stimulated cAMP accumulation was high as compared with NK3R–CHO cells (Nakajima et al., 1992).
