2022, Journal of Molecular BiologyCitation Excerpt :Both the ADRA2C and the CXCR3 genes encode Gi protein-coupled receptors, being adrenoceptor alpha 2c (α2C-R) activated by catecholamines (e.g., epinephrine or norepinephrine),35 and c-x-c motif chemokine receptor 3 (CXCR3) by chemokines CXCL9, CXCL10 or CXCL11.36 Since receptor stimulation leads to the inhibition of adenylyl cyclase (AC) and consequently blocks the synthesis of the second messenger cAMP from ATP,37 we can infer that knocking down these genes (and consequently inhibiting Gi) can increase intracellular cAMP levels. On the other hand, receptor overexpression could lead to AC inhibition, thus lowering intracellular cAMP levels ([cAMP]i).
