Bufalin for an innovative therapeutic approach against cancer
2022, Pharmacological ResearchCitation Excerpt :Their biosynthesis takes place in the adrenal gland and cholesterol is the major substrate [5]. Bufalin-like immunoreactive substances have been observed in human serum for several years [11], but the presence of bufalin in human serum was validated for the first time in 2020 by HPLC-MS/MS [12]. This study reported bufalin concentrations of about 5.7 nM in the serum of healthy individuals and of five times decreased concentrations in the serum of patients with hepatocellular carcinoma, further highlighting its role as a potentially protective molecule against cancer development.
Endogenous cardiotonic steroids and cardiovascular disease, where to next?
2020, Cell CalciumCitation Excerpt :It is likely that other bufadienolides may also be present in circulation. Indeed there are reports of proscillaridin A- [47] and bufalin-like [48] immunoreactive substances detected in human serum. Some of the studies have relied on the use of antibody-based ELISA methods for detection of the endogenous CTS whereas others have used high-resolution mass spectrometry and NMR, though often without detailed validation.
Simultaneous determination of seven bufadienolides in rat plasma after oral administration of Shexiang Baoxin Pill by liquid chromatography-electrospray ionization-tandem mass spectrometry: Application to a pharmacokinetic study
2014, Journal of Chromatography B: Analytical Technologies in the Biomedical and Life SciencesCitation Excerpt :Therefore, an integral investigation into the PK evaluation of the bufadienolides in SBP could not only link data from pharmacological assays to clinical efficacy/toxicity, but also help exploring rational dosage regimens. Various analytical methods based on liquid chromatography technology have been developed for the determination of the bufadienolides in biological fluids [18–21]. However, high limit of quantification, long analysis time, lack of commercial assay kit, the costliness of the instrument and strict requirement for sample pretreatment or could not fulfill oral PK study due to the insufficient sensitivity and specificity, though pronounced.
Quantitative determination of arenobufagin in rat plasma by ultra fast liquid chromatography-tandem mass spectrometry and its application in a pharmacokinetic study
2013, Journal of Chromatography B: Analytical Technologies in the Biomedical and Life SciencesCitation Excerpt :However, the use of arenobufagin in pharmacokinetic studies has not been reported. HPLC/UV [10–13,9], fluoroimmunoassay [14], and enzyme immunoassay [15,16] methods have been developed to analyze toad venom by determining bufadienolides, bufalin, cinobufagin, and resibufogenin in biosamples. These methods are not sensitive enough or are very expensive, which limits their application.
