Biphenyl-indanones: Allosteric potentiators of the metabotropic glutamate subtype 2 receptor

Abstract

We have identified and synthesized a series of biphenyl-carboxylic acid indanones as allosteric potentiators of the metabotropic glutamate receptor 2. Structure–activity relationship studies directed toward improving the potency and the brain to plasma ratio of the initial lead led to the discovery of 5 and 23 (EC50 = 111 and 5 nM, respectively).

The metabotropic glutamate receptor 2 has been implicated in a number of central nervous system disorders. Herein, we report on the discovery, synthesis, and biological evaluation of biphenyl-indanones as potent small molecule mGluR2 allosteric modulators.

      The literature survey revealed that various methods have been developed for the synthesis of indanones [33–37]. Indanone and its derivatives can be used as useful intermediates in the synthesis of (R) and (S)-2–hydroxy–indanone [38], 6-substituted 3,4-benzotropolones [39], dimer of 2-(4-pyridylmethyl)-1-indanone [40], biphenylcarboxylic acid indanones [41] and 1-indanone thiosemicarbazone derivatives [42]. The critical intermediate 4-chloro-2-(2-fluoroethyl)-5-methoxy-2,3-dihydro-1H-inden-1-one used for the total synthesis of (7β-9aβ)-1,4-dichloro-2-hydroxygibba-1(10a),2,4,4b-tetraen-6-one [43].

      The narrow linker connecting the upper and lower chambers in mGlu5 rationalises the predominance of an alkyne linker or equivalent bioisostere (e.g. MPEP, dipraglurant and fenobam; docked poses shown in Figure 4a — see also Figure 2b).       Interestingly, CBiPES was more easily dissolved than BINA under our conditions, and while both drugs were entirely dissolved for injection, this cannot be excluded as a possible explanation of the difference in efficacy between these two PAMS. Both of these compounds have been characterized as selective mGluR2 PAMS (Ahnaou et al., 2009; Benneyworth et al., 2007; Bonnefous et al., 2005; Fell et al., 2010; Galici et al., 2006; Johnson et al., 2005, 2003; Sanger et al., 2012).
