Interplay between cholesterol and drug metabolism
2011, Biochimica et Biophysica Acta - Proteins and ProteomicsNovel CAR-mediated mechanism for synergistic activation of two distinct elements within the human cytochrome P450 2B6 gene in HepG2 cells
2005, Journal of Biological ChemistryCitation Excerpt :Our present work revealed that the corresponding CYP2B6 region does not play a role in the OA synergistic response and did not significantly affect induction by TCPOBOP alone. In addition, deletion of the region corresponding to the CYP2B1/2-positive element identified by Padmanaban and co-workers, to which protein binding was enhanced by OA or PB in rat (24–26), did not alter either the TCPOBOP-induced or the OA synergistic activity of the CYP2B6 gene. Moreover, the sequence of the OARE did not resemble those of the previously identified enhancer elements.
Phenobarbitone-mediated translocation of the cytosolic proteins interacting with the 5′-proximal region of rat liver CYP2B1/B2 gene into the nucleus
2002, Biochemical and Biophysical Research CommunicationsA conserved nuclear receptor consensus sequence (DR-4) mediates transcriptional activation of the chicken CYP2H1 gene by phenobarbital in a hepatoma cell line
2000, Journal of Biological ChemistryCitation Excerpt :These findings explain why CYP2H1 mRNA is induced by OA to a certain level and that this OA induction is not changed when adding PB. Our results may explain why in the rat, OA inhibitsCYP2B1/2 mRNA induction by PB but induces aCYP2B1/2 minigene containing the proximal sequence of the 5′-flanking region in reporter gene assays (39, 49). The similarity of these data to the inhibition of the transfer of CAR to the nucleus in mouse liver suggests that a CAR-like receptor protein is involved in induction mediated by the 264-bp PBRU in the CYP2H15′-flanking region of the chicken.
