The nature of receptors regulating the formation of cyclic AMP in brain tissue

Abstract

Extensive studies of the past seven years have provided fundamental information on the pharmacological properties of receptors controlling formation of cyclic AMP in brain slices and homogenates. Receptors for adenosine, α- and β-adrenergic agonists, dopamine, serotonin, H₁- and H₂-histaminergic agonists, and prostaglandins of the E series have been defined and evidence for a glutamate receptor has been presented. Extrapolation of such pharmacological data to studies with whole animals should provide important information as to the physiological significance of specific cyclic AMP-generating systems to the function of the intact brain.

Adenosine inhibition of the regeneration in vitro of adult frog sciatic sensory axons

The sensory axons of the adult frog sciatic nerve have earlier been shown to regenerate in vitro. If a local test crush is made at the initiation of culturing, regeneration starts after 3.4 days and proceeds at a rate of about 0.8–0.9 mm/day for several days. In the present experiments regeneration was inhibited by adenosine in a reversible and dose-dependent fashion. Similarly, both an adenosine analogue, 2-chloroadenosine (2-CA), and a non-hydrolyzable ATP analogue, AMP-PNP, reduced the outgrowth of sensory axons. The effect of adenosine was partially antagonized by theophylline at a critical concentration. Using a compartmental system, it could clearly be shown that adenosine exerted its effects at the outgrowth region. Adenosine, 2-CA, and AMP-PNP were also found to inhibit the proliferation of Schwann cells in the regenerating nerve. Various experiments showed that the latter can not explain the outgrowth inhibitory effects, which could be mediated by adenosine receptors associated with the elongating axons.
Noradrenergic potentiation of cerebellar Purkinje cell responses to GABA: evidence for mediation through the β-adrenoceptor-coupled cyclic AMP system

Previous in vivo studies from our laboratory have consistently shown that iontophoretically applied norepinephrine (NE) can potentiate γ-aminobutyric acid (GABA)-induced depressant responses of cerebrocortical, cerebellar and hypothalamic neurons. Additional experiments have further suggested that this noradrenergic facilitating action is specific for GABA and results from the activation of a β-type adrenoceptor. The goal of the present studies was to determine if the cAMP second messenger system might also be a component of the mechanism responsible for this NE modulatory action on GABA-mediated inhibition. In one set of in vitro experiments, we examined cerebellar neuronal responses to GABA before, during and after iontophoretic application of NE, 8-bromo3′,5′-cylic AMP (BcAMP) or 3-isobutyl-1-methyl xanthine (IBMX) or bath application of forskolin (10–30 μM). In a second group of in vivo studies, extracellularly recorded responses of individual cerebellar Purkinje (P) cells to iontophoretic pulses of GABA or β-alanine were examined before, during and after NE or BcAMP microiontophoresis. In 20 of 25 cerebellar cells recorded from tissue slices, iontophoretically applied NE markedly enhanced responses to GABA in a manner similar to that observed previously in vivo. In these in vitro preprarations, bath application of forskolin was also capable of potentiating GABA-induced inhibition in each of 4 cases tested whereas dideoxy-forskolin was not. Iontophoretic application of IBMX further enhanced the facilitating effects of NE on GABA-induced inhibition in 10 of 11 cases tested. Furthermore, under in vitro conditions, BcAMP augmented inhibitory responses to GABA in all cerebellar neurons tested. In the intact rat brain, iontophoretic administration of BcAMP caused a marked NE-like augmentation of P-cell responses to GABA in 73% of the cells tested. As with NE, BcAMP was ineffective in enhancing P-cell inhibitory responses to β-alanine, an agent which like GABA causes hyperpolarization, by increasing Cl⁻ conductance. In summary, these results indicate that a membrane permeant analog of cAMP, a phosphodiesterase inhibitor and an agent which directly activates adenyl cyclase can mimic the previously observed GABA-potentiating actions of NE. Thus, these findings provide further support for the contention that noradrenergic enhancement of GABA inhibition results from a cascade of transmembrane events which includes β-receptor activation, adenyl cyclase stimulation and increased intracellular production of cAMP.
Neurochemical and neuropharmacological aspects of histamine receptors

Histamine H₁ and H₂ receptors have been identified in the mammalian central nervous system. Both receptor subtypes have been indirectly characterized by a variety of receptor-mediated biological assays and directly through the utilization of radioligand binding techniques. Two types of psychotherapeutic agents, the antidepressants and neuroleptics, are potent inhibitors of H₁ and H₂ receptors. However, most compounds tested have a higher affinity for H₁ receptors. Like many other neurotransmitter receptors, H₁ receptor function can be regulated by guanine nucleotides, various ions, and by the degree of agonist stimulation.
Alpha-receptor-mediated facilitation of somatosensory cortical neuronal responses to excitatory synaptic inputs and iontophoretically applied acetylcholine

The major objective of the present study was to characterize, in terms of α- and β-receptor mechanisms, the previously observed facilitating actions of norepinephrine (NE) on somatosensory cortical neuronal responses to excitatory synaptic inputs and iontophoretic administration of acetylcholine (ACh). In the forelimb region of rat somatosensory cortex (SI), excitatory single unit responses to natural stimulation of the contralateral forepaw or microiontophoretic pulses (10 sec duration at 45 sec intervals) of ACh were examined before, during and after iontophoretic administration of NE, phenylephrine (PE) and isoproterenol (ISO). Adrenergic agonist actions were quantitatively assessed by computer-based analysis of poststimulus time or cholinergic response histograms. At doses which suppressed or had no effect on the spontaneous discharge, the α agonist, phenylephrine, facilitated excitatory synaptic transmission in 12 of 14 cells and enhanced neuronal responsiveness to ACh in 15 of 24 cells. Thus, phenylephrine consistently mimicked the potentiative actions of NE. whereas the β agonist, isoproterenol, at similar doses, was ineffective in enhancing synaptic efficacy (n = 12) or ACh-induced excitation (n = 19). In addition, iontophoretic application of the α blocker, phentolamine. reversibly antagonized NE-induced potentiation of ACh responses in 6 out of 6 cases. In contrast, administration of the beta antagonist, sotalol, had no effect on NE-mediated enhancement of ACh in all three neurons tested. Overall, these results suggest that endogenously released NE may facilitate excitatory synaptic transmission within the somatosensory cortex by activation of postsynaptic adrenoceptors with α characteristics.

Effects of adenosine and related compounds on an inhibitory process in rat cerebral cortex

Inhibition of neurons in the rat cerebral cortex was evoked by local cortical stimulation. Adenosine, AMP, and ATP applied by microiontophoresis produced no change of this inhibition. Theophylline and aminophylline, administered intravenously or iontophoretically, blocked the depression of neuronal firing by adenosine, but did not themselves affect the duration of inhibition. Dipyridamole and hexobendine, at iontophoretic doses which potentiated responses to adenosine, produced some reduction of inhibitory duration. We conclude that endogenous purines do not normally contribute to local cortical inhibition, but that their accumulation can inhibit this phenomenon, possibly presynaptically.
