2021, Neuroscience and Biobehavioral ReviewsCitation Excerpt :Relatively to its distribution in the enteric nervous system, SP is mainly localizable in the pylorus, gastric fundus, duodenum, and jejunum (Sternini et al., 1989). SP binding its NK1-R in the gut muscular layers, including the longitudinal and circular muscles, as well as the muscularis mucosae, regulates the contractile activity of the entire gastrointestinal tract (Holzer-Petsche, 1995). Beside the gastrointestinal localization, SP synthesis and transport through the vagus nerve toward the brain are demonstrated in humans and other mammals (Brimijoin et al., 1980; Funakoshi et al., 1989; Gamse et al., 1979; Katz and Karten, 1980; Lundberg et al., 1979; MacLean, 1987).

SP, NKA and NKB were shown to have high affinity to neurokinin (NK) type-1 receptor (NK-1R), NK type 2 receptor (NK-2R), and NK type 3 receptor (NK-3R), respectively (Regoli et al., 1994; Holzer-Petsche, 1995; Maggi, 1995; Kurtz et al., 2002; Lecci et al., 2006). In study in vitro, we recently demonstrated that NK-2R mRNA was abundantly expressed in the omasal muscle layers and that application of NKA elicited potent tonic contraction of omasal muscle strips in sheep (Onaga et al., 2012).

In general, tachykinin NK1 and NK2 receptor agonists contract smooth muscle, and tachykinin NK3 receptor agonists modulate motility via receptors located on enteric neurones (Holzer-Petsche, 1995; Shimizu et al., 2008). Studies using S. murinus isolated ileum have demonstrated contractile responses to nicotine, acetylcholine, histamine (Takamura et al., 1988), and 5-hydroxytryptamine (Javid and Naylor, 1999a,b; Takamura et al., 1988), and relaxant responses to noradrenaline and adrenaline (Takamura et al., 1988).
