Abstract

Three dopamine receptor mRNAs were detected by reverse polymerase chain reaction in the rat forebrain during ontogeny. The D₁ receptor mRNA gave a faint signal from gestational day 11 (G11) on, while the D_1b receptor was not detectable before G12. The D₃ receptor mRNA was slightly expressed at G11 and clearly present at G14. Our results show that these mRNAs are present at early stages of fetal life, each one with its specific time of appearance.

Chapter IX Human forebrain dopamine systems: Characterization of the normal brain and in relation to psychiatric disorders

Impairments of dopaminergic neuronal populations have been strongly implicated in a variety of neurological, psychiatric and drug addiction disorders, mental illnesses. Although most research efforts have been directed toward understanding the dopamine neural system in animals, technological advances over the past two decades have also helped to expand our current knowledge about the discrete anatomical organization of the dopamine system in the human brain. Dopamine neuronal populations have now been characterized in the human brain from the level of gene transcription (using in situ hybridization histochemistry) to the distribution of related proteins (immunohistochemistry) and their receptors (as visualized in vivo by neuroimaging techniques of positron emission tomography (PET) and single photon emission tomography (SPECT). The insights revealed about the human dopaminergic system by these technologies should help in the development of more targeted treatment interventions of neuropsychiatric disorders. In this chapter, we have summarized the current status regarding the neuroanatomical organization of the human dopamine systems with focus on the forebrain. The general organization of the dopamine system, from the level of synthesis, release, uptake and receptors, in the human brain are described with their potential implications for addiction and psychiatric disorders.
Developmental expression of the zinc finger transcription factor DRRF (dopamine receptor regulating factor)

Dopamine receptor regulating factor (DRRF) is a novel transcription factor with unique anatomical distribution and functional properties, suggesting its importance in regulating dopaminergic neurotransmission. To gain insight into the in vivo function of this factor during embryogenesis, we studied its distribution at embryonic days E8–E16 in the mouse using in situ hybridization. DRRF mRNA is expressed uniquely during development at all time points tested with high levels observed at E12, E14 and E16 in various tissues. DRRF expression is also found in particular brain regions, such as the neopallial cortex, olfactory lobe and corpus striatum. This pattern of DRRF distribution during embryogenesis overlaps with that found in the adult brain, and with the expression profile of dopamine receptors both in the adult and during development.
Catecholamine systems in the brain of vertebrates: New perspectives through a comparative approach

A comparative analysis of catecholaminergic systems in the brain and spinal cord of vertebrates forces to reconsider several aspects of the organization of catecholamine systems. Evidence has been provided for the existence of extensive, putatively catecholaminergic cell groups in the spinal cord, the pretectum, the habenular region, and cortical and subcortical telencephalic areas. Moreover, putatively dopamine- and noradrenaline-accumulating cells have been demonstrated in the hypothalamic periventricular organ of almost every non-mammalian vertebrate studied. In contrast with the classical idea that the evolution of catecholamine systems is marked by an increase in complexity going from anamniotes to amniotes, it is now evident that the brains of anamniotes contain catecholaminergic cell groups, of which the counterparts in amniotes have lost the capacity to produce catecholamines. Moreover, a segmental approach in studying the organization of catecholaminergic systems is advocated. Such an approach has recently led to the conclusion that the chemoarchitecture and connections of the basal ganglia of anamniote and amniote tetrapods are largely comparable. This review has also brought together data about the distribution of receptors and catecholaminergic fibers as well as data about developmental aspects. From these data it has become clear that there is a good match between catecholaminergic fibers and receptors, but, at many places, volume transmission seems to play an important role. Finally, although the available data are still limited, striking differences are observed in the spatiotemporal sequence of appearance of catecholaminergic cell groups, in particular those in the retina and olfactory bulb.
Behavioral responses to a D<inf>1</inf> dopamine agonist in weanling rats treated neonatally with cocaine and Δ<sup>9</sup>-tetrahydrocannabinol

We determined whether neonatal exposure to cocaine with or without Δ⁹-tetrahydrocannabinol (THC) altered the behavioral responses of weanling rats to the full D₁ dopamine (DA) agonist SKF 81297. Rats were injected SC once daily from postnatal day (PD) 1 through 5 with cocaine (20 mg/kg), the same dose of cocaine plus THC (10 mg/kg), or drug vehicle. On PDs 24, 25, or 26, male and female littermates were administered 3 or 10 mg/kg of SKF 81297 or saline vehicle, and then tested 15 min later in an open-field apparatus. Neither neonatal drug treatment nor gender influenced the behavioral responses to SKF 81297. The drug challenge did, however, produce several dose-dependent behavioral effects, including increases in locomotor activity, line crossing, sniffing, and headshakes, and a decreased incidence of rearing, grooming, and stationary behavior. Furthermore, even though earlier administration of cocaine and THC failed to alter D₁ receptor sensitivity, animals in both neonatal treatment groups exhibited an overall increase in grooming behavior and a decrease in sniffing compared to controls when the results were combined across doses of SKF 81297. These findings indicate that early postnatal exposure to cocaine can alter certain behaviors independently of functional changes in the D₁ receptor system.
Partial purification of a pramipexole-induced trophic activity directed at dopamine neurons in ventral mesencephalic cultures

We previously demonstrated that media conditioned by exposure to ventral mesencephalic (VM) cultures in the presence of pramipexole (PPX) and other drugs with dopamine (DA) D₃ properties, increased the growth and survival of DA neurons in recipient VM cultures. This trophic activity was heat-labile and not present in parietal cortex cultures or cultures pretreated with the DA neuron toxin MPP⁺. In an effort to begin to identify the protein(s) responsible for this trophic effect, we compared the conditioned media from normal VM cultures, VM cultures incubated with PPX, and VM cultures pretreated with MPP⁺ and treated with PPX. Neutralization studies using anti-GDNF and anti-BDNF failed to reduce the conditioned media transfer effect, and Millipore Ultrafree centrifugation studies placed the mol.wt. of the activity around 30 kDa. SDS separation revealed three potential bands of interest. A 35-kDa band was present in normal cultures, increased in PPX-incubated cultures, and absent in MPP⁺-pretreated/PPX-incubated cultures. This conforms to the effect the protein concentrates used to produce these gels had on the growth of DA neurons in VM cultures. Since VM cultures grown in neural basal media, which inhibits the growth of glia, still responded to PPX in a dose-dependent fashion, the trophic activity may be a DA autotrophic factor. However, the gels also revealed two bands at ∼31 and 55 kDa that were reduced by exposure to PPX and present in MPP⁺-pretreated cultures. The possibility that these are neuroinhibitory factors that are also regulated by PPX therefore cannot be ruled out.
The locomotor effects of a putative dopamine D<inf>3</inf> receptor agonist in developing rats

Dopamine receptors have been categorized into subfamilies D₁ and D₂, each with separate roles in dopamine-mediated behaviors. Of the D₂ subfamily, the dopamine D₃ receptor has been cloned, but the behavioral effects of selectively stimulating the D₃ receptor are largely unknown. The purpose of this study was to quantify the locomotor responses of developing rats to the putative dopamine D₃ receptor agonist, 7-hydroxy-N,N-di-n-propyl-2-aminotetralin (7-OH-DPAT). One of three doses of 7-OH-DPAT (0.01, 0.10, 1.00 mg/kg) or saline was injected subcutaneously into rats at the age of 10, 20, 30, or 60 days. Five minutes after the injection, rats were placed in automated activity monitors which recorded locomotor behavior at 5 min intervals for 2 h. The high dose of 7-OH-DPAT increased locomotor activity in rats of all ages. The medium and low doses increased activity in 10- and 20-day-old rats but not in 30- or 60-day-old rats. The level of drug-induced activation peaked at 20 days of age. In 30- and 60-day-old rats, but not 10- and 20-day-old rats, a period of locomotor suppression preceded the activation in response to the high dose of 7-OH-DPAT. In rats aged 20 days and older, the middle and low doses decreased locomotion early in the test session, but activation did not ensue. This dose-response pattern across ontogeny closely resembles that induced by quinpirole, an agonist at the dopamine D₂ receptor subfamily.
