The AVPR2 gene (located on Xq28) encodes a protein of 371 amino acids (Birnbaumer et al., 1992), which belongs to the G protein coupled hormone receptor (GPCR) family. AVPR2 has seven membrane-spanning helices, an extra cellular domain that binds AVP, and an intracellular domain that activates the Gs protein via its third intracellular loop i3 and part of its proximal C-terminal region to induce the renal antidiuretic pathway (Birnbaumer et al., 1992; Birnbaumer, 1992). Like most GPCRs, AVPR2 contains a conserved pair of extracellular cysteine residues that link the first and second extracellular loops via a disulfide bond (Fig. 3).

Ach stimulated the muscarinic receptors followed by stimulation of phospholipase C (PLC) activity and increases the [Ca2+]i and protein kinase C (PKC) activity (Selbie and Hill, 1998). The μ receptor stimulated by morphine and then activated Gβγ subunits play an important role in signal transduction process (Birnbaumer, 1992). It is widely accepted that Gβγ is the signal transducer for pertussis toxin (PTX)-sensitive PLC stimulation found for many Gi-coupled receptors (Camps et al., 1992).

AC is stimulated by a variety of physiologically relevant extracellular stimuli, such as Prostaglandin E2 (PGE2), through G-protein (Gs)-coupled membrane receptors [12]. In addition, AC can be activated by pharmacological agents, such as forskolin (FSK), which is a direct activator of AC [13], isoproterenol (ISO), which is a synthetic agonist for the β-adrenergic family of receptors [14], or cholera toxin (CTX), which causes constitutive activation of Gs by stimulating ADP-ribosylation of its α-subunit [15]. Protein kinase A (PKA) is the most important effector of cAMP action, although other cAMP-binding proteins, such as cAMP-regulated guanine nucleotide exchange factors (cAMP-GEFs), can mediate some of the biological functions of cAMP in a PKA-independent manner [16].

Spot 14 was identified as a type of G protein, and spot 22 as a cGMP-dependent protein kinase. These proteins may participate in G protein-coupled receptor-mediated signal transduction pathway or cGMP-mediated signal transduction pathway (Birnbaumer, 1992; Leonard, 2001). In conclusion, DA has an effect on SL-1 cell proliferation and apoptosis.
