2006, Brain ResearchCitation Excerpt :Together with the reversal effect of PPADS, a nonselective antagonist of P2X-receptor (Chizh and Illes, 2000; North and Surprenant, 2000), it can be deduced that the actions of αβ-meATP on cutaneous fibers are mediated by P2X3, P2X1 and/or P2X2/3 receptors. Although all of these receptors may contribute to αβ-meATP-evoked afferent discharges, the widespread distribution of P2X3 in small-diameter DRG neurons and the low expression of P2X1 mRNA in DRG suggest that P2X3 is the dominant receptor for αβ-meATP in sensory neurons (Barden and Bennett, 2000; Dunn et al., 2001; North and Surprenant, 2000; Waldron and Sawynok, 2004). In a recent study, it has been shown that only a small number (39%) of CMH and AMH fibers were activated by the maximal dose of αβ-meATP (5 mM) in vitro (Hamilton et al., 2001).

For example, clusters of P2X1 receptors of about 0.4–1 μm in diameter have been observed by immunocytochemistry on the smooth muscle cells of the vas deferens, bladder, arteries, as well as on sensory neurons of the dorsal root ganglia and sympathetic neurons of the superior cervical ganglion (Barden et al., 1999; Dutton et al., 1999; Hansen et al., 1999; Barden and Bennett, 2000; Li et al., 2000; Pulvirenti et al., 2000; Liang et al., 2001). P2X1 receptors can also form clusters when expressed in Xenopus oocytes and HEK293 cells (Dutton et al., 2000).

The expression pattern of P2X3 receptors in sensory ganglia has also been studied by immunohistochemistry at both the light microscope (Vulchanova et al., 1997, 1998; Bradbury et al., 1998; Xiang et al., 1998a; Novakovic et al., 1999; Barden and Bennett, 2000) and electron microscope (Llewellyn-Smith and Burnstock, 1998) levels. In DRG, intensive P2X3 immunoreactivity is found predominantly in a subset of small- and medium-diameter neurons, although absent from most large neurons.
