2015, Ageing Research ReviewsCitation Excerpt :Many studies have found increased sensitivity to the effect of benzodiazepines in older people in the absence of any attributable pharmacokinetic change (Kanto et al., 1981; Albrecht et al., 1999), whereas for some benzodiazepines no changes in pharmacodynamic response have been observed (Pomara et al., 1997). The exact mechanism of these pharmacodynamic changes for benzodiazepines are not well established; GABA receptor binding itself is not affected by the ageing process (Ruano et al., 1996; Sundman et al., 1997; Bickford and Breiderick, 2000). Since most of these pharmacodynamic studies have examined older patients only, the altered disease pathophysiology in older patients (compared to younger patients) may be a reason for the observed changes in the pharmacodynamic behaviour of medications (McLean and Le Couteur, 2004).

As with human studies, there have also been some investigators who have not found an increased sensitivity to benzodiazepines in aged rodents (Bickford and Breiderick, 2000; Wikinski et al., 2001). Additional studies have addressed receptor interactions of benzodiazepines, which bind at a specific site located on the GABAA receptor.
